Одной из причин генетической нестабильности индуцированных плюрипотентных стволовых клеток (ИПСК) является репликационный стресс (РС). РС приводит к формированию разрывов и пробелов хроматид в районах конститутивных ломких сайтов (кЛС), которые являются «горячими точками» для формирования хромосомных перестроек. Процесс митотическогоб синтеза ДНК (MiDAS) служит для восстановления целостности генома при РС и является маркером кЛС.</p></sec><sec><title>   Цель и задачи</title><p>   Цель и задачи: картирование хромосомных разрывов и сайтов MiDAS в ИПСК.</p></sec><sec><title>   Методы</title><p>   Методы. Индукцию РС проводили афидиколином и кофеином. Районы MiDAS детектировали по включению EdU на ранних этапах митоза. Дифференциальное окрашивание проводили с помощью DAPI и актиномицина Д.</p></sec><sec><title>   Результаты</title><p>   Результаты. Проанализированы 3 линии ИПСК (P1L5, P12L3, P16L1). Cреди 800 картированных хромосомных разрывов было выявлено 33 кЛС. Охарактеризованы как описанные ранее, так и уникальные для ИПСК кЛС. Выявлены кЛС, ко-локализованные с точками разрыва рекуррентных аномалий кариотипа ИПСК.</p></sec><sec><title>   Выводы</title><p>   Выводы. Впервые описан репертуар кЛС в ИПСК и их ассоциации с рекуррентными перестройками.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>   Introduction</title><p>   Introduction. One of the causes of genetic instability in iPSCs is replication stress (RS). RS leads to the formation of chromatid breaks and gaps in regions of constitutive fragile sites (cFS), which are «hotspots» for chromosomal rearrangements. The process of mitotic DNA synthesis (MiDAS) serves to restore genome integrity under RS and acts as a marker of cFS.</p></sec><sec><title>   Objective</title><p>   Objective: mapping chromosomal breaks and MiDAS sites in iPSCs.</p></sec><sec><title>   Methods</title><p>   Methods. RS was induced using aphidicolin and caffeine. MiDAS regions were detected by EdU incorporation at early mitotic stages. Differential staining was performed using DAPI and actinomycin D.</p></sec><sec><title>   Results</title><p>   Results. Three iPSC lines (P1L5, P12L3, P16L1) were analyzed. Among 800 mapped chromosomal breaks, 33 cFS were identified.
