Одним из условий достоверности и воспроизводимости исследований на клеточных моделях является их генетическая стабильность. Для индуцированных плюрипотентных стволовых клеток (ИПСК) характерно спонтанное возникновение генетических аномалий. Контроль генетической стабильности ИПСК необходим каждые 5-10 пассажей в процессе культивирования. Оптимизация подходов к рутинному анализу генетической стабильности ИПСК требует изучения спектра аномалий, характерных для этих клеток, их структуры и потенциальной функциональной значимости.</p></sec><sec><title>   Цель</title><p>   Цель: выявление и анализ аномалий генома в культурах ИПСК, полученных в российских лабораториях.</p></sec><sec><title>   Методы</title><p>   Методы. Линии ИПСК (n = 66), полученные в пяти лабораториях из материала 6 здоровых доноров и 27 пациентов с диагностированными генетическими заболеваниями (15 нозологий), были проанализированы методом кариотипирования. Обнаруженные аномалии кариотипа валидировали методами флуоресцентной гибридизации in situ, спектрального кариотипирования и хромосомного микроматричного анализа.</p></sec><sec><title>   Результаты</title><p>   Результаты. Аномалии кариотипа были выявлены в 23,6 % образцов и включали трисомию по хромосомам 20 и 8, изохромосому 20q и структурные перестройки с участием хромосом 1, 2, 8, 15, 18. Рекуррентные увеличения числа копий сегментов хромосом 1q и 20q составили 69 % выявленных аберраций.</p></sec><sec><title>   Выводы</title><p>   Выводы. Структура и частота аномалий кариотипа в выборке линий ИПСК из российских лабораторий сопоставима с ранее опубликованными данными, полученными при анализе линий из зарубежных коллекций. Высокая доля рекуррентных аберраций указывает на перспективность внедрения таргетных методов анализа генетической стабильности клеточных линий.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>   Background</title><p>   Background. Genetic stability of model cell cultures is a fundamental prerequisite for ensuring the validity and reproducibility of experimental results. Induced pluripotent stem cells (iPSCs) are inherently prone to developing spontaneous genetic abnormalities. Thus, monitoring the genetic stability of iPSCs every 5–10 passages during culture expansion is essential. Optimizing methods for routine analysis of iPSC genetic stability requires an in-depth investigation of the spectrum of abnormalities characteristic of these cells, along with their structural features and potential functional implications.</p></sec><sec><title>   Aim</title><p>   Aim: to identify and analyze genomic anomalies in iPSC cultures derived from Russian laboratories.</p></sec><sec><title>   Methods</title><p>   Methods. iPSC lines (n = 66), derived in five laboratories from biological material sourced from six healthy donors and 27 patients diagnosed with genetic diseases (15 nosologies), were analyzed using karyotyping techniques. Identified karyotype anomalies were validated through fluorescent in situ hybridization, spectral karyotyping, and SNP microarray analysis.</p></sec><sec><title>   Results</title><p>   Results. Karyotype abnormalities were observed in 23.6 % of the samples, including trisomy of chromosomes 20 and 8, isochromosome 20q, and structural rearrangements involving chromosomes 1, 2, 8, 15, and 18. Recurrent gains of chromosome segments 1q and 20q accounted for 69 % of the detected aberrations.</p></sec><sec><title>   Conclusions</title><p>   Conclusions. The structure and frequency of karyotype abnormalities in iPSC lines derived from Russian laboratories are consistent with previously reported data from studies of iPSC lines in international collections.
