В исследовании принимали участие 226 добровольцев, подписавших информированное согласие, в числе которых 65 пациентов с новообразованиями хориоидеи. По данным клинико-инструментальных методов обследования все больные, в свою очередь, были разделены на две группы – опытную (пациенты с увеальной меланомой и прогрессирующим невусом хориоидеи, n=42) и группу риска (пациенты со стационарным невусом хориоидеи, n=23). Контрольную группу (n=61) составили лица без внутриглазных новообразований. В популяционную выборку вошли лица русской национальности (согласно проведенному анкетированию), случайным образом выбранные в г. Москве. Генотипирование по полиморфизмам гена ZNF350 (rs2278415, rs2278420 rs4986773) выполнено методом ПЦР с последующей рестрикцией ДНК.</p></sec><sec><title>Результаты</title><p>Результаты. При сравнении частот аллелей и генотипов по полиморфным локусам rs2278415 и rs498677 гена ZNF350 статистически значимых различий между изученными группами не выявлено. В контрольной группе генотип AA и аллель А по полиморфизму rs2278420 гена ZNF350 обнаруживались значимо реже, чем в опытной группе, группе риска и популяционной выборке.</p></sec><sec><title>Заключение</title><p>Заключение. В ходе проведенного исследования показано, что генотип АА и аллель А по полиморфному локусу rs2278420 гена ZNF350 ассоциированы с увеальной меланомой, прогрессирующими невусами хориоидеи и стационарными невусами хориоидеи. Также более высокие частоты указанных генотипа и аллеля обнаружены в популяционной выборке, что может быть связано с эффектом «случайной выборки» и отсутствием данных об отягощенном онкологическом семейном анамнезе.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Aim</title><p>Aim: to investigate the association of ZNF350 (rs2278420, rs2278415, rs4986773) gene polymorphisms with the risk of uveal melanoma development.</p></sec><sec><title>Methods</title><p>Methods. The study involved 226 volunteers who signed an informed consent, including 65 patients with UM and choroidal nevi (study group). According to the results of the clinical and instrumental examination, all patients were divided into two groups – a study group (patients with uveal melanoma and progressive choroidal nevus, n=42) and a risk group (patients with benign choroidal nevus, n=23). The control group (n=61) included individuals without intraocular neoplasms. The population group included Russian individuals (according to the survey data) randomly selected in Moscow. Genotyping for the ZNF350 rs2278415, rs2278420, and rs4986773 gene polymorphisms was carried out by PCR followed by DNA restriction.</p></sec><sec><title>Results</title><p>Results. Comparing the allele and genotype frequencies for the ZNF350 rs2278415 and rs4986773 gene polymorphisms, no statistically significant differences were found between the studied groups. In the control group, the genotype AA and allele A for the ZNF350 rs2278420 gene polymorphism were determined significantly less frequently than in the study, risk, and population groups.</p></sec><sec><title>Conclusion</title><p>Conclusion. Our study has revealed, the AA genotype and A allele at the polymorphic locus rs2278420 of the ZNF350 gene are associated with UM, progressive, and benign choroidal nevi.
