Целью настоящего исследования было изучение ассоциаций полиморфизма плейотропных генов с БА и её эндофенотипами. Проанализировано 23 полиморфных варианта 17 генов. Выявлены ассоциации с астмой и её эндофенотипами девяти генов (IL12A, LTA, TNF, TNFRSF1B, IFNGR2, NOS3, AGTR1, GNB3, PPP3R1); показано, что в развитие изолированной БА и её сочетаний с сопутствующими заболеваниями ассоциированные гены вовлечены в различной степени. Продукты большинства генов, для которых установлены ассоциации с БА и её эндофенотипами, участвуют в реализации двух основных механизмов патогенеза БА: воспаления и бронхоспазма. Установлено, что клинические фенотипы лёгкой, средней и тяжёлой форм БА имеют свои отличительные генетические особенности. Для генов PPP3R1, TNFRSF1B, GNB3, AGTR1 ассоциативное исследование с БА проведено впервые.</p></abstract><trans-abstract xml:lang="en"><p>Bronchial asthma is clinically heterogeneous multifactorial pathology with genetic component. Association of 23 polymorphic variants in 17 genes with asthma and asthma endophenotypes was studied. The association for the 9 genes (IL12A, LTA, TNF, TNFRSF1B, IFNGR2, NOS3, AGTR1, GNB3, PPP3R1) with asthma and asthma related phenotypes has been shown. Most gene products associated with bronchial asthma and asthma related phenotypes are involved in realization of two basic pathways of asthma pathogenesis, i.e. inflammation and bronchial spasm.
